I wouldn't suggest that as a sensible alternate route to get to the Keys, unless you want to stop in Miami on your way South.
You could even head East from Orlando and head all the way down the east coast. If you headed east from Orlando and came south on I95 you'll not really see anything much more than you would on the Turnpike, I don't think. Unless you want to spend a lot of time detouring off the interstate into places and exploring, or there's anywhere you really want to see, I'd just head off early and get down here. I would either just hit the Turnpike and maybe use the time saved to go into the Everglades when you get down here, or use Using 27 will add about an hour or so vs the Turnpike in my limited experience.
Coming down to Key Largo to then making day trips back up to The Everglades, and more so Miami, will add a lot of driving, and you could most likely spend less on hotels around the Miami area vs in Key Largo also.

That is the Saturday of the big parade in Key West Fantasy Fest , when something like 60, will be driving south to attend.
